What is DNA fragmentation?

Each sperm carries in its head DNA with the genetic information necessary for fertilization and healthy development of the embryo. When this DNA is damaged or “torn”, we speak of fragmentation. It can be the result of:

  • Oxidative stress;
  • Inflammatory processes;
  • High temperature (varicocele, work in a hot environment);
  • Aging of spermatozoa;
  • Harmful habits (smoking, alcohol, drugs);
  • Bad lifestyle and stress.

Why is the Halosperm test important?

In men with normal spermograms but with fragmented DNA:

  • The chance of natural conception is lower;
  • Fertilization may occur, but the embryo stops developing;
  • The risk of miscarriage is increased;
  • It is possible that the embryos look good morphologically but are genetically unstable.

How is the test performed?

  1. Collection of semen sample — after 2—5 days of sexual abstinence;
  2. Sample processing in the laboratory;
  3. Halosperm staining — a “halo” is created around the head of normal spermatozoa;
  4. Microscopic analysis — hundreds of sperm are counted to determine the percentage of DNA fragmentation.
